Lisa Raymond and Mike Bryan were the defending champions, but lost in Quarterfinals to Cara Black and Wayne Black.

Katarina Srebotnik and Bob Bryan won the title, defeating Lina Krasnoroutskaya and Daniel Nestor 5–7, 7–5, [10–5] in the final. It was the only US Open and 2nd Mixed Doubles title for Srebotnik, and the 1st Mixed Doubles title for Bryan, in their respective careers.
